qPCR kit that allows detecting Porcine Reproductive and Respiratory Syndrome Virus (PRRSV) vaccine strains (MLV, ATP, Fostera) or related strains (vaccine-like strains) in various samples (serum, lungs, oral fluids, etc.).

DESCRIPTION:
This kit is a 3-plex qPCR assay that allows detecting and differentiates the 3 vaccine strains of Porcine reproductive and respiratory syndrome virus (PRRSV): MLV-, ATP- and Fostera-like vaccine strains

SUMMARY AND EXPLANATION:
Porcine reproductive and respiratory syndrome virus (PRRSV) is one of the most important swine pathogens. Virus eradication programs frequently use mass vaccination with modified live vaccines. In North America three modified live vaccines are widely used i.e. Ingelvac PRRS MLV and ATP (Boehringer Ingelheim) and Fostera PRRS (Zoetis).
The monitoring of PRRSV eradication programs relies on regular PCR testing of samples such as serum, oral fluid or processing fluids. However PCR results interpretation is complicated since positive results may be due to field or vaccine virus. This results in additional costs and time to sequence positive cases in order to differentiate field and vaccine virus. In addition sequencing is not always successful as the virus load may be below the sensitivity level of sequencing.
In order to address these issues we have developed a 3-plex real time PCR assay (qPCR) which allows rapid, sensitive, and specific detection and differentiation of the 3 vaccine strains in various samples.

Biovet Swinecheck® PRRSV MLV-ATP-FOS qPCR contains primers and probes for the identification and quantification of MLV-, ATP- and Fostera-like vaccine strains. The kit provides every component of the qPCR reaction (buffers, enzyme, dNTPs, specific primers and probes for the pathogen and an endogenous amplification control).
MLV-, ATP-, and Fostera-like virus genome amplification is detected through the FAM, VIC and TAMRA channels, respectively. A positive control including synthetic MLV, ATP and Fostera specific DNA is provided for relative and absolute quantification.
The assay was validated using StepOne™ and 7500 FAST Real Time PCR Systems (Applied Biosystems®), but the kit is expected to run in a wide range of Real Time PCR thermal cycler machines.
