The Xpert miRNA cDNA Synthesis Kit provides an efficient and fast method for the synthesis of high quality first-strand microRNA cDNA from total RNA templates. First-strand cDNA can be directly used as template in (q)PCR, using the included miRNA Reverse prime, which binds to the incorporated oligo(dT)-adapter sequence and an adequate miRNA specific forward primer.
GENERAL INFORMATION:he Xpert miRNA cDNA Synthesis Kit provides an efficient and fast method for the synthesis of high quality first-strand microRNA cDNA from total RNA templates.
The Xpert miRNA cDNA Synthesis kit comprises Poly(A) Polymerase, which catalyzes the addition of adenosine residues to the 3´-ends of RNA molecules (polyadenylation), and an oligo(dT) that anneals to the newly formed poly(A)-tail in order to start cDNA synthesis by the included Xpert Reverse Transcriptase an (RNase H-) RNA-dependent DNA polymerase.
Besides Poly(A) polymerase, Oligo(dT), and Xpert RTase, the Xpert miRNA cDNA Synthesis Kit also contains an RNase inhibitor for protection of template degradation, reaction buffer, and dNTPs. GRiSP’s RNase inhibitor has an improved resistance to oxidation in comparison with human RNase inhibitors and is stable under even very low concentrations of DTT, making it an excellent choice for RNA protection.
Finally, the Oligo(dT) present in the reaction mixture is linked to a specific adapter sequence, which is complementary to the Xpert miRNA Reverse primer (included). First-strand cDNA can be directly used as template in qPCR, using this miRNA Reverse primer and an adequate Forward primer that is specific for the microRNA under study.
