INTENDED USE:The IGF-1 600 ELISA is a manual enzyme immunoassay for the quantitative measurement of Insulin-like Growth Factor 1 (IGF-1) in human serum. For in vitro diagnostic use. For laboratory professional use. The device is intended to be used as an aid to diagnosis for individuals where information on one or more of the following is required: differential diagnosis of growth disturbances especially in children; acromegaly; impaired growth; dwarfism; chronic liver disease & hepatocellular dysfunction.

GENERAL INFORMATION: Insulin-like growth factor 1 (IGF-1), also called somatomedin C, is a single-chain 70-amino acid polypeptide, that bears structural similarity to insulin (1,2). The IGFs (IGF-1 and IGF-2) are growth factors produced in the majority of the organs and body tissues mainly by the regulation of pituitary growth hormone. They have autocrine, paracrine and endocrine actions on metabolism and cell proliferation, growth and differentiation (3-5). Approx. 98% of circulating IGF-1 are bound with high specificity and affinity to a family of 6 IGF binding proteins (IGFBP 1-6) that modulate their bioactivity. IGFBP-3 is thought to be the major binding protein of IGF-1, accounting for around 80% of all bound IGF-1, typically forming a tri-molecular complex of 150 kDa with IGF-1 and an acid-labile subunit. Insulin-like growth factor-2 (IGF-2) is a protein closely related to IGF-1 and capable of binding to the GF-1R as well as its own IGF-2 receptor (IGF-2R). However, whilst the IGF-1R is capable of signal transduction, the IGF-2R is not and binding results in lysosomal degradation of IGF-2. This provides an indirect method of IGF-1 regulation by regulating the level of IGF-2 that is available to bind to the IGF-1R (6-7). Most of the known IGF actions are mediated via IGF type 1 receptor (IGF1R). IGF-1 concentrations change with age, nutritional status, body composition and growth hormone secretion. Vitamin D has been shown to increase circulating IGF-1 and IGFBP-3.
