Detection of Alaska pollack (Gadus chalcogrammus) DNA in food products and animal feedstuff. For use in real time instruments with FAM and ROX detection channels.
BACKGROUND: According to the European Commission Directive 2002/86/EC, food ingredients must be declared. Additionally, according to the EU Directive 1379/2013, the fish species must be disclosed on the label with both commercial and scientific denominations in order to verify the origin and traceability of the fish and to analyze the quality control of handling and cleaning processes of production lines.
Moreover, the species authenticity is also highly relevant to the consumer for diverse reasons such as economic, health, cultural, and religious. Fraudulent substitution of cheaper meats/fishes in place of more expensive ones, the inclusion of meat in non-meat/fish (vegetarian) products, and the interest of knowing the animal speciation in meat products (halal and kosher) are clear examples for the importance of this issue.
Several methods for species detection are based on the identification of proteins by means of electrophoretic and/or immunological methods. However, these methods are not reliable for application in highly processed and heated products due to protein deterioration. DNA is more stable than proteins during processing and although it can be fragmented by several processes, DNA methodologies like PCR based techniques still allow the identification of DNA from the different species present in a sample. Real time PCR techniques, using specific fluorescent probes, are especially suitable for these products because small fragments of DNA can still be amplified and identified with high sensitivity and specificity.
PRODUCT DESCRIPTION: SUPREME REAL TIME DETECTION Alaska pollack (Gadus chalcogrammus) uses real-time PCR for the detection of Alaska pollack (Gadus chalcogrammus) DNA present in the mitochondrial genome in a simple, reliable, and rapid procedure. The assay is based on 5’ nuclease real time PCR reactions to amplify a unique genomic sequence in the target. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the target as well as a positive control and an internal control (IC). To minimize PCR cross-contamination,the Kit contains dUTP and uracil-N glycosylase (UDG). Comparative quantification analysis of DNA in food and feedstuff products to estimate the percentage of the fraudulent products is also possible using in combination with the DNA Standard SUPREME Alaska pollack (Gadus chalcogrammus) Kit (optional).
